Somatic cells can be reprogrammed to induced pluripotent stem (iPS) cells by expression of defined transcription factors. However, only a small percentage of the cells end up as fully reprogrammed. Moreover, in many attempts to generate iPS cells, partially reprogrammed novel cell types are obtained, often bearing the proliferative features of iPS cells but lacking full pluripotency. The reprogramming process can thus serve as a model system for studying cell fate decisions in mammalian cells. We examine the early dynamics of transcription-factor mediated reprogramming through live-cell imaging, probing when and how the decision to commit to an iPS fate is made.
